In search of selective proteases tuned to the substrate of your need.

Proteases are used by all organisms such as animals, plants, fungi and bacteria. They regulate all kinds of biological functions including cell signaling, blood clotting, digestion or apoptosis (programmed cell death). The latter make them important candidates for targeting cancer.

The selective cleavage of specific peptide bonds by proteases is pivotal for normal cell function. A good example is for instance the regulation of pro-insulin to insulin, and many other hormones. But also in research selective proteases are widely used for protein identification or processing of tagged proteins.

Endoprotease services aims to develop a selective protease platform for any desired substrate. Using site selective mutagenesis in combination with computational design and machine learning, we are able to adapt the protease to the desired sequence. We can change the substrate pocket to fit any amino acid side-chain, even non-natural ones. The focus of our research is on subtilisins, which are already widely used to modify antibodies and give access to antibody drug conjugates. But also in the detergent industry as well as food and feed, subtilisins are broadly applied.
